A study led by Dr Alexi Nott at UK DRI at Imperial finds that different brain-cell types are linked to genetic risk for dementia in distinct ways.

The cell type risk genes we identified were mostly disease specific indicating unique vascular and immune mechanisms across these pathologies. Lastly, we integrated gene expression analysis to prioritise repurposable drugs with genetic evidence for the treatment of AD.

Using nanoCUT&Tag epigenomic profiling and pcHiC chromatin conformation analysis we found AD genetic risk is mostly immune with some vascular enrichment, whereas small vessel disease, which is often comorbid with AD, is strongly enriched for blood vessel cell types.

The brain neurovascular epigenome and its association with dementia Ziegler et al. reveal distinct genetic and epigenetic mechanisms underlying Alzheimer’s disease and cerebral small vessel disease within neurovascular cells, identifying immune-related AD risk and broad neurovascular involvement in SVD. They highlight novel regulatory pathways and potential therapeutic targets, advancing understanding of these comorbid conditions for future treatment strategies.
