The implications are profound, and go beyond TLR7/8: TLR9 uses the same basic design to discriminate self/nonself DNA. Together, TLR7/8/9 must be the key system to distinguish between retroviral particles and normal exosomes: evolution HAD to come up with a way to do this.

2β€²-O-Methyl-guanosine RNA fragments antagonize TLR7 and TLR8 to limit autoimmunity - Nature Immunology Here, the authors show that 2’-O-methylated RNA fragments from ribosomal RNA naturally block TLR7 and TLR8, helping to avoid avoid harmful self-RNA detection and autoimmunity.

How does your immune system tell pathogenic RNA from your own, even though they’re made of the same stuff?
Our latest paper with @gantierlab.bsky.social shows some self RNA act as an off switch preventing autoimmunity in the absence of pathogens. πŸ§¬πŸ”¬β

I am delighted to share our characterisation of natural antagonism of TLR7/8 in Nature Immunology. We propose a model where 2’O-Methyl RNA fragments keep TLR7/8 in check, preventing autoimmunity. > www.nature.com/articles/s41...

IsomiR stoichiometry changes as disease biomarkers. Gantier, Gearing and colleagues demonstrate, through analysis of small RNA sequencing data, that quantification of microRNA isoforms enables identification of accurate biomarker candidates overlooked in aggregated microRNA data. A ratio-based approach to isoform-level biomarker discovery is presented, measuring the relative abundance of alternate isoforms to provide internal normalisation within a microRNA.

Gomez-Diaz, @v-hornung.bsky.social et al. show that mice lacking RNase T2 display an inflammatory phenotype in a TLR13-dependent manner, even under germ-free conditions, suggesting that RNase T2 plays an important role in limiting self-RNA recognition https://buff.ly/4jG49Gy
