Updated architectural model of the NPC from our lab, starred by bsky-less Agnieszka Obarska-Kosińska. Using cryo ET, XL-MS and modeling, we identify novel nucleoporins, among those the mRNA export complex TREX-2! Check out the preprint on biorxiv #TeamMassSpec #TeamTomo

Metabolically regulated proteasome supramolecular organization in situ Structural steps along the assembly of proteasome storage granules—membraneless organelles that form in response to metabolic shifts in yeast—are visualized inside cells by cryo-electron tomography. Inactive 26S proteasomes oligomerize into trimers, which assemble into paracrystalline arrays that serve as reservoirs of fully assembled proteasomes under conditions of low energy, ready for reactivation when glucose is restored.

LLOMe has long been used to study lysosomal damage, yet how it works has remained a mystery.
Using cryo-electron tomography, we show it forms amyloid structures inside lysosomes that mechanically rupture membranes – revealing a new paradigm for lysosomal failure.

This one was quite the journey! The paper describing the #ChlamyDataset is finally out and on the cover of Mol Cell!

This beautiful rendering made by co-author @jessheebner.bsky.social and Holly Peterson shows an instance of mitochondrial fission found in the dataset 😍

[Maybe long thread ahead]

Good guess, but Chlamy lacks 'canonical vaults'. My speculation for the lightbulb complex would be one of the five name-less Chlamy SPFH proteins (www.ebi.ac.uk/interpro/ent...), or maybe Prohibitins, though they’re usually IMM-localized in other organisms.

Take advantage of this fantastic opportunity to become part of the SCALE community! Several professorships and group leader positions are available at Goethe University and FIAS. We are looking for professors in the fields of molecular microbiology, cellular biochemistry, and molecular biochemistry.

It’s out! Using cryo-ET in Dicty cells, we take a fresh in situ look at vaults. Surprisingly, we uncover vaults associated with ER and NE membranes, and find that many vaults enclose ribosomes in defined orientations, opening new avenues to their cellular function! www.biorxiv.org/lookup/conte...

Just published! We share an open-source workflow to measure membrane thickness from tomograms, including a tutorial with 3D visualizations. We analyze thickness variations across organelles and reflect on where to define a membrane boundary. @becklab.bsky.social #teamtomo rupress.org/jcb/article/...

The 3-decade-old competition that enabled the emergence and evaluate of AlphaFold has run out of NIH funding.

The program will be terminated in weeks.

This is terminating success.
