A brief T–cell–mediated rejection occurred early, likely due to residual lymph node immunity, but was successfully treated. No antibody-mediated rejection was seen.

🐷➑️🧬 In the first pig-to-human kidney transplant, a multiomics approach demonstrated that persistent innate immune activation (monocytes/macrophages) remains a key barrier. Better control of innate immunity will be critical for future success.

Discovery from MGB Researchers Yields Insights on Preventing Organ Transplant Rejection | Mass General Brigham The study identifies receptors that slow inflammation, signaling a promising target for new therapies.

πŸ’‘ Takeaway: Innate immune checkpoints like Siglec-E/7/9 are emerging as crucial regulators of transplant rejection. 4/4

βœ… Blocking NF-ΞΊB or TNF-Ξ± in Siglec–E–deficient models restores graft survival.

βœ… In humans, higher expression of SIGLEC7/9 (homologs of Siglec-E) is linked to better allograft outcomes β€”highlighting their potential as biomarkers or therapeutic targets. 3/4

βœ… Siglec-E suppresses dendritic cell activation, limiting NF-ΞΊB signaling and TNF-Ξ± production.

βœ… Loss of Siglec-E accelerates rejection in murine heart, kidney, and skin transplants through heightened T cell–mediated immunity. 2/4

The inhibitory receptor Siglec-E controls antigen-presenting cell activation and T cell–mediated transplant rejection Siglec-E negatively regulates innate immune responses and acute T cell–mediated transplant rejection in mice, offering a potential therapeutic target.

🚨 Our new study in @SciTranslMed identified the inhibitory receptor Siglec-E as a critical brake on dendritic cell activation and T cell–mediated transplant rejection. 1/4
