Don't be fooled by imidazole! It absorbs UV280, just like protein β on chromatogram it'll be seen! Imidazole is great for purifying His-tagged proteins, but it's a pain for when you want to detect and measure concentration of proteins using UV280 absorbance! youtu.be/JZeTMXfOtGU

Bands basically appear instantaneously without background and you can save and reuse multiple times. I used it in grad school and am so glad I got some again!
