Our results show that itaconate biology in human macs differs from mouse and that this metabolite has distinct functions in the macrophage populatons that Mtb infects during aerosol infection.

We hope this iPSC derived system will be useful for studying AM biology in diverse contexts.

Irg1 expression in myeloid cells prevents immunopathology during M. tuberculosis infection | Journal of Experimental Medicine | Rockefeller University Press Nair et al. define a key role for Irg1 in minimizing the pathological immune response associated with Mtb infection. Using Irg1โˆ’/โˆ’ and Irg1fl/fl conditiona

2. Itaconate has distinct functions in macrophage subtypes. In AMs, there is a loss of bacterial control in irg-/-, but In blood derived human Macs, itaconate negatively regulates the Mtb induced type I interferon and TNF responses, similar to what @stallingslab.bsky.social reportedin mouse BMDMs

We made IRG1/ACOD1 deficient iPSCs via gene editing, produced different macrophage types, and examined their phenotypes with LPS and Mtb infection. Major conclusions:

1. Human Macs produce substantially less itaconate than mouse macrophages, and AMs produce even less

28.07.2025 21:43 โ€” ๐Ÿ‘ 1    ๐Ÿ” 0    ๐Ÿ’ฌ 1    ๐Ÿ“Œ 0

We then used the genetic tractability of iPSCs to derive macrophages deficient in IRG1/ACOD1, which makes the metabolite itaconate. Itaconate has complex effects in mouse macrophages. However, the human ACOD enzyme is less active than mouse:

To faciliate human AM studies, we derived macrophage subtypes from iPSCs under distinct cytokines, including GM-CSF, which is required for development of AMs.

Alveolar macrophages (AMs) are the first cell of contact for many inhaled pathogens, including M. tuberculosis, are developmentally distinct from blood macrophages, are difficult to study in humans due to the necessity of bronchoscopy, and are not genetically tractable.

Macropinocytosis maintains CAF subtype identity under metabolic stress in pancreatic cancer Zhang et al. demonstrate that macropinocytosis inhibition induces myCAF-to-iCAF conversion through a metabolically stressed, MEK-ERK-dependent intermediate state. This CAF plasticity remodels the pancreatic tumor microenvironment and improves responses to immunotherapy and chemotherapy, supporting macropinocytosis inhibition as a promising approach for combination treatment strategies in pancreatic cancer.
