We hope this iPSC derived system will be useful for studying AM biology in diverse contexts.
2. Itaconate has distinct functions in macrophage subtypes. In AMs, there is a loss of bacterial control in irg-/-, but In blood derived human Macs, itaconate negatively regulates the Mtb induced type I interferon and TNF responses, similar to what @stallingslab.bsky.social reportedin mouse BMDMs

1. Human Macs produce substantially less itaconate than mouse macrophages, and AMs produce even less
We then used the genetic tractability of iPSCs to derive macrophages deficient in IRG1/ACOD1, which makes the metabolite itaconate. Itaconate has complex effects in mouse macrophages. However, the human ACOD enzyme is less active than mouse:

We show that these iPSC derived alveolar macrophages, which we call AM-L (like), resemble bonafide AMs at baseline and with Mtb infection, leveraging the insights of our collaborators in the Hawn lab published here:

To faciliate human AM studies, we derived macrophage subtypes from iPSCs under distinct cytokines, including GM-CSF, which is required for development of AMs.
