Our data suggest that targeting epithelial PPAR-α may be a strategy to reverse lipotoxicity and restore epithelial homeostasis in UC. We also show patient-derived colonoids are a viable model for testing future epithelial-directed therapies. Full paper: rdcu.be/eT2x8

Excitingly, pharmacological inhibition of PPAR-α successfully reprograms epithelial metabolism in aUC colonoids. This inhibition suppressed hypermetabolism, limited lipid accumulation, shifted energy use towards glucose, and reduced chemokine secretion and cytotoxicity. //6

Jide identified a master lipid regulator, PPAR-α, as a major factor driving this metabolic dysfunction. Furthermore, activating PPAR-α in control colonoids was sufficient to induce lipid-related hypermetabolic stress, similar to what we observed in UC organoids. //5

This inefficient hypermetabolic phenotype is driven by lipid dysregulation! aUC colonoids showed excess lipid accumulation and were dependent on fatty acid oxidation (FAO) to sustain their high oxygen consumption rates (OCR) //4

Jide found colonoids from active UC patients exhibit hypermetabolism and cellular stress, especially during differentiation. OxPhos is inefficient, with high proton leak, oxidative stress, cytotoxicity, and chemokine production. //3

UC therapies overwhelmingly target the immune system, yet only 30% achieve mucosal healing. We need strategies focusing on the dysfunctional epithelium. We addressed this by developing patient-derived colon organoids (colonoids) from pediatric UC patients as preclinical model //2
