Paradoxically, one of these factors, HLA-B*35 is associated with a rapid progression to AIDS if the treatment is not initiated. It is truly exciting that early ART initiation might have transformed an initial desfavorable situation into an enhanced potential to achieve HIV remission.

Statin treatment by itself will not be sufficient to promote HIV control in the context of ART interruption, but CD8 T cell in vivo metabolic reprogramming with rosuvastatin could be used to potentiate the efficacy of other immunotherapies aiming at HIV cure or remission

We identified several changes in the survival and functionality of CD8 T cells that coincided with rosuvastatin treatment. These parameters went back to baseline levels upon cessation of the treatment.

In our new study we show that CD8 T cell reprogramming is also achievable in vivo. We used bio-banked samples from the CESAR trial where people on ART (without metabolic disorders but subpotimal immune reconstitution) received 3 months of rosuvastatin treatment to decrease activation/inflammation.

We then showed that, targeting metabolic pathways, it was possible to reprogram in vitro CD8 T cells from people receiving antiretroviral treatment so that they acquire some of the properties observed in the cells from HIV controllers
