02.11.2025 13:12 β π 1 π 0 π¬ 1 π 0Thank you so much!!
01.11.2025 19:18 β π 1 π 0 π¬ 0 π 0I also want to point out that this work was entirely funded by NIH through the HEAL Initiative. The future of HEAL is somewhat uncertain right now and I hope this paper will play a small role in convincing folks that this work is important and will have an impact on pain science and treatment.
01.11.2025 12:26 β π 1 π 0 π¬ 0 π 0Thanks for reading my thread! Doing this work has been a dream come true for me. I am incredibly grateful to Katherin Gabriel and @oliviadavis.bsky.social for making this amazing journey possible and to the organ donors and their family for the gift of knowledge they have given us.
01.11.2025 12:24 β π 1 π 0 π¬ 1 π 0Finally, we compared mice to humans and saw conservation of most neuronal types, but divergences in neuronal proportions and transcriptomes, in particular for cell surface receptors. If you want to do drug discovery, these details matter!!
01.11.2025 12:23 β π 1 π 0 π¬ 1 π 0We then went to town on the glial populations where we identified some pretty profound sex differences. We did not note sex differences in neurons, but the identification of male and female specific glial populations for astrocytes, microglia, and oligodendrocytes could have important functions.
01.11.2025 12:22 β π 0 π 0 π¬ 1 π 0And, of course, we did the same with the ventral horn, giving confidence in our identification of motor neuron pools.
01.11.2025 12:20 β π 0 π 0 π¬ 1 π 0We used this information to create detailed maps of the locations of cell types in the dorsal horn. This gives great insight into sensory/nociceptive circuits in the human dorsal horn with a resolution that has not been seen before at this scale.
01.11.2025 12:19 β π 1 π 0 π¬ 1 π 0Of course, we also care about non-neuronal cells and the spatial arrangement of all of these cell types so we used Xenium spatial transcriptomics to map the locations of these cell types using gene markers from our single nucleus work. Our custom panel led to many important insights.
01.11.2025 12:17 β π 0 π 0 π¬ 1 π 0We did the same thing for ventral horn neurons (we microdissected the dorsal horn from the ventral horn to have clear sequencing) and were able to clearly delineate motor neuron populations.
01.11.2025 12:15 β π 0 π 0 π¬ 1 π 0We mapped dorsal horn neurons using single nucleus sequencing and developed detailed transcriptomes for subtypes which we named using a nomenclature system based on work done at the Allen Brain Atlas.
01.11.2025 12:13 β π 1 π 0 π¬ 1 π 0We did single nucleus sequencing and Xenium @10xgenomics.bsky.social spatial transcriptomics on male and female spinal cords from organ donor recovered tissues in collaboration with Southwest Transplant Alliance. We started this work about 5 years ago. We optimized and optimized and optimized.
01.11.2025 12:12 β π 1 π 0 π¬ 1 π 0The latest work from our PRECISION Human Pain Center project is now up on @biorxivpreprint.bsky.social led by Katherin Gabriel and @oliviadavis.bsky.social with a huge contribution from @allanhpool.bsky.social's lab and, of course, the Southwest Transplant Alliance: www.biorxiv.org/content/10.1...
01.11.2025 12:10 β π 20 π 10 π¬ 4 π 0Also huge props to @allanhpool.bsky.social
and his lab at UTSW for helping optimise our single nuc prep and looking for species differences between human and mice.
This paper builds on the foundation already set and increases our understanding of human spinal cord at an even higher resolution.
Thrilled to share our latest preprint using single nuq sequencing and Xenium to define neuron organisation and sex differences in glia within human spinal cord. Truly a collaborative effort, with a big shoutout to Katherin Gabriel and @tedpricethepainguy.bsky.social

We have a great Department and UTD is still a great place to work!
31.10.2025 21:15 β π 1 π 0 π¬ 0 π 0We are seeking a senior tenured professor in our Department working on affective and/or motivational neuroscience (defined in the ad, but pretty broad). The idea is that this person is going help lead our growth in this area alongside our excellent Centers. jobs.utdallas.edu/postings/30693

28.10.2025 22:57 β π 7 π 0 π¬ 0 π 0wonder if we could convince them to chant "Fire Nico"?
27.10.2025 20:22 β π 1 π 0 π¬ 0 π 0Sharing a figure from our recent paper showing excitatory synapses in the human spinal dorsal horn. Iβll never get over the awe of seeing these tiny dots, which underly such an important part of the human experience!
