1.) be patient getting started takes time
2.) every time in the 1st 6mo a colleague asks to get coffee do it.
3.) it’s a marathon not a sprint.

Pretty cool. With help from some friends modified a 3D printer to make a fraction collector. It cost $200$ instead of 1500$ works perfectly.

In addition the "in-cell" spectra look to clean to truely be in-cell. Now i have never done RNA in-cells but As a Pielak PHD and Gronenborn Postdoc who did in-cell NMR in each lab I can comment that these spectra are very suspect to me.

look at these supernatant controls...they saw leakage in their post and just scrolled down in intensity to try to flatten it out. I have NEVER seen an NMR spectrum without resonances look that much like a flat line and then on top if you look at where the deviations from flat correlate with peaks

This work of course was done by two talented members of the Guseman Lab, Sabrina Richter our former all star tech who just left us for grad school at UC-Berkeley and it was started by the Neal Brook during their summer rotation last year. I am sure this is the first of many stories from them!

Our data suggests that the glycocalyx could be able to modulate the energy landscape between these states and thus influence cell signalling. Given the dynamic nature of the glycocalyx and its ability to remodel itself as response to stimuli/cell stress. This is an exciting ave for future study!

These results demonstrate the glycopolymer components of the glycocalyx can tune stability and interactions of proteins by acting as macromolecular crowders. Contextually, many of the proteins that reside in the glycocalyx are involved in signalling and exist in inactive state A and active state B.

Switching next to protein protein interactions, we used arguably my favorite protein A34F GB1 to demonstrate that these glycopolymers stabilize protein protein interactions in a similar fashion to protein folding, via chemical interactions.
