In vitro experimental techniques can provide detailed information about specific properties of mechanosensitive ion channels. However, an experimental gap remains in understanding how channels are activated and the consequences of their function in complex in vivo settings.

In their Opinion article, Samantha Webster @llssam.bsky.social, Remi Brynn and Kate Poole @katepoole.bsky.social consider how perspectives from both cell biologists and biophysicists will be crucial for characterising mechanosensitive ion channels in vivo.
