Context-dependent requirements for Foxp3 in #Treg functions. Foxp3 is required for function of newly generated Tregs, but Foxp3 loss in mature Treg cells resulted in minimal functional changes under steady state. However, tumoral Tregs are uniquely sensitive to loss of Foxp3.

Local delivery of IL-15 and anti-PD-L1 nanobody by in vitro-transcribed circILNb elicits superior antitumor immunity in cold tumors Niu et al. construct the circCV-B3 vector to enable scarless circRNA engineering. Utilizing circCV-B3 vector, they engineer circILNb for co-encoding IL-15 and anti-PD-L1 nanobody. Additionally, they demonstrate circILNb’s potent suppression of both local and distal tumors. The authors also demonstrate the superior antitumor efficacy of circILNb compared to protein-based therapeutics.
