Our findings reveal a unique splice variant and a trapped substrate intermediate never observed before, offering new insights into proteolytic catalysis. We also show proteasomes cluster from the post-meiotic spermatid stage, with striking human-mouse differences in proteasome organisation!

We discovered giant membrane-less islands of proteasomes in human sperm nuclei, containing 20S and 20S-PA200 populations! We solved their in situ structure at sub-nanometer resolution (up to 6.8 Γ…) and in vitro at near-atomic resolution (1.8 Γ…)!

We discovered that the SUN5-Nesprin3 LINC complex forms a hexagonal lattice at the sperm HTCA, which radically reshapes the nuclear membrane locally!
Our findings also provide the structural basis for understanding the severe ASS (Acephalic Sperm Syndrome) male infertility phenotype.
