Now in my independent lab we are working on a range of different phosphoinositide conversions. From PI(5)P to PI(4,5P2 conversion during peroxisome membrane contact site formation to the PI(3)P to PI conversion by the Myotubularins during receptor trafficking.

By segmenting the beads we can quantify these differences and show that active Arf6 strongly enhances the reaction speed of phosphoinositide conversion by the PIP5K. This has drastic effect on membrane trafficking pathways! www.cell.com/current-biol...

Importantly it also allows us to directly compare beads with different surface functionalisation. Here I attached the constitutively active mutant of the small GTPase Arf6 (Arf6-QL) and the inactive mutant to beads and we see complete separate reaction kinetics!
