Identifying Effective Cryoprotectant Agents for Emerging Bacterial Model Species Host-associated bacteria live amongst eukaryotes within varied niches and form relationships ranging from facultative to obligate. With advancement in studies of such symbiotic associations, fastidious bacteria are increasingly becoming targets for genetic manipulation. However, there are limited resources for screening possible agents enabling in vitro culturing and storage of these microbes. In this study, we present a simple protocol for optimizing cryopreservation of non-model organisms in laboratory settings using conventional chemicals. Our initial motivation for this observation was to discover a cryoprotection agent for independently cultured Mycetohabitans spp., a fungal endosymbiont. We tested several common bacterial cryoprotection agents like glycerol, bovine serum albumin (BSA), and dimethyl sulfoxide (DMSO) over an ultra-low freeze-thaw cycle to determine an adequate method of cryoprotection for assorted bacteria. We observed different recovery rates across bacterial species and cryopreservation methods, and identified cryoprotectants that reliably resulted in viable bacteria for each of the strains tested. We present this as a resource for those working with other fastidious and host-associated bacteria that may be missing effective cryopreservation methods. ### Competing Interest Statement The authors have declared no competing interest.

I've been curious about alternative cryoprotectants in -80C since even. The typical Ralstonia are divas about freeze/thaw. I've considered trying sucrose and trehalose as alternatives to glycerol.

From reading the older literature and the genome reduction, I suspect the clove strains might survive stresses less effectively. Try to store/ship them in multiple formats (water, PBS, broth media, agar stabs).

It's the weird outlier in our figure without the expected eps orthologs. NCBI RefSeq flagged that small genome strain as too small because their pipeline is likely looking for a typical RSSC genome. It's small even for a clove strain, but I am curious.

I am really curious about the natural variation of that lineage. The original publications from Eden-Green and colleagues describe a mix of mucoid and non-mucoid colony morphologies. One of the available genomes is extremely small (~3.5 Mb) and lacked the eps gene cluster by BLAST.
