We have cloned several of this size for replicative (low copy) in gram negative but integrative in gram positives. The homolog arms and double antibiotic resistance cassettes makes them quite big though. Might be worth considering integrase mediated genome integrations for these purposes.

We discovered a tiny archaeon with the smallest known archaeal genome — only 238 kbp!
Candidatus Sukunaarchaeum mirabile has almost no recognizable metabolic pathways and may rely heavily on a host to survive.
It also represents a novel, deep-branching lineage in the archaeal tree.

Wonder which would take longer: Mediaformulation or the actual reduction of the genome.

1/2 New paper by our PhD student Sompot Antimanon, where he uses Pseudomonas putida as a biocatalyst for cinnamic acid production in a process that increase water use efficiency. Sompot reached a TRY of T=143 g/L, R= 2.25 g/L/h, and Y 1.50 g/g glc after 8 recycles.
