Up close and personal with a branching neurite in a DIV2 hippocampal neuron expressing FTractin-GFP. This was imaged every 6 seconds over 10 minutes on the trusty @zeiss-microscopy.bsky.social LSM880. Happy #FluorescenceFriday
Also looks like it has some actin within it as well. You've really got me wondering now π
Although the final frame looks like it may have some attachment, or just a coincidental new branch forming in the same area.
That's a great question, I have no idea lol. I'm only collecting one z-plane here so its possible if its a neuronal structure I could be missing its attachment to the neighboring neurite.

Imaging neurons quickly for 3min typically won't give you huge cell-wide cytoskeletal events, but you do get to capture fun little tugs and loops in the soma and neurites (as well as whatever the hell is going on in the bottom right).

I do most of my troubleshooting imaging with NBM/B27 but if I need good clear imaging I would use NEUMO/SOS. I'll have to check but I believe the longest I did with that media was overnight and it was definitely reduced autofluorescence. Relatively new, now relatively old I guess.
Following since I've just been informed NEUMO/SOS is discontinued π§
The world desperately needs more microscope merch. Iβd spend half my stipend.
