Thank you for the compliment. Full transparency: the cartoon was AI-assisted โ€” but it required serious scientific mentoring ๐Ÿ˜‚ Convincing ChatGPT that anaphase = sister chromatids, not X-shaped chromosomes was a multi-cycle process. Peer review works, even for AI!

Congratulations, @ltovini.bsky.social! What a journey this has been! This story would not exist without your determination, resilience, and scientific imagination. You kept pushing when things were difficult and turned complexity into clarity. I am super proud of you!!

Thanks for the nice words! Indeed... yeast could be seen as an exception (as they keep transcription mostly on) but not all over :D also needs to be off at some sites. And there are even more interesting things on Pol I that will come on another upcoming ms. Stay tuned ;)

5/ This means that mitotic transcription isnโ€™t just a passive consequence of cell cycle stage โ€” it actively threatens genome stability when not properly regulated.

4/ The result? When transcription isnโ€™t cleared, sister chromatid resolution is impaired โ€” a critical step needed to ensure faithful chromosome segregation in anaphase.

3/ Importantly, we show that these defects arise because R-loops form when transcription persists during mitosis โ€” and inhibiting transcription reverts the defects. That establishes a causal link.

2/ Using TTF2 depletion to force nascent transcripts to persist on mitotic chromatin, we found that this abnormal retention leads to mitotic errors including abnormal chromosomes and bridges.

1/ During mitosis, cells globally suppress transcription as chromosomes condense โ€” a conserved hallmark of cell division. But what if this repression fails? ๐Ÿ˜ฎโ€๐Ÿ’จ

๐Ÿงต Thread: What happens when transcription isnโ€™t properly shut off during mitosis? Our new preprint explores just that ๐Ÿ‘‡

It was an honour to present at such a special meeting and get to know other recently elected EMBO members (and โ€œolderโ€ too). Many thanks to Helder Maiato for the nomination and to all members and staff for the warm welcome.
