These discoveries originated from our finding that TOFA, a drug widely used to inhibit de novo lipogenesis, also inhibits protein S-acylation. This is a word of caution to other researchers using this drug, as it may inhibit many proteins using fatty acyl-CoA as substrate!

Healthy hematopoietic stem/progenitor cells in contrast have high intrinsic mitochondrial #flexibility, readily switching to other nutrients when glutamine breakdown is blocked. This shows that leukemia cells handle metabolic stress differently, creating a therapeutic opportunity!

Protein S-acylation is a post-translational modification in which fatty acids are covalently bound to proteins. When #glutamine catabolism is blocked, increased S-acylation of #mitochondrial proteins with long saturated fatty acids preserves metabolic health and leukemia cell survival.

While AML cells express EAAT1, the transporter was not needed for AML growth or survival after #chemotherapy, neither in cell cultures nor in a mouse model. Our results further indicate that AML cells possess multiple mechanisms to acquire #aspartate, underscoring the importance of this #metabolite!
