In summary we found that Hang regulates transcription at the level of chromatin modification. It limits local H4K16ac deposition by NSL, thereby preventing overproduction of mRNA. We hypothesize that this fine-tuning of stress genes might play a role in the development of ethanol tolerance.

To address this, we studied the NSL-catalyzed histone mark H4K16ac in Hang knock-out cells. We found that H4K16ac increases at a fraction of TSSs upon Hang loss. The underlying genes are derepressed upon Hang depletion but recover their repressed status upon further removal of NSL.

We further investigated the interplay between Hang, a repressor, and the NSL complex, an activator of transcription. Both show robust interaction with each other (Co-IP). However, they have opposite effects on gene expression (RNA-seq upon RNAi). How can this be reconciled?

We showed that Hang binds to the transcriptional start sites of actively transcribed genes (ChIP-seq) and dampens their expression (RNA-seq upon RNAi). We think that this might protect cells from overshooting transcription of highly active genes.

Using a CRISPR/Cas9-based approach in a Drosophila cell line coupled with mass spectrometry, we found that Hang interacts with a broad spectrum of chromatin-regulating protein complexes (e.g. dREAM, Ino80, Sin3, dMec, TFIID, NSL).
