For some actual process videos of these kinda paintings, I can reccommend ニッカー絵具【公式】NICKER COLOUR CO.,LTD's The Joy of Painting series (where you see the traditional techniques for anime BGs) or 手ジタルラボ　kazutube, who does original and reproduction backgrounds in digital.

I'd assume this particular shot they layed down the city lights from below first, then painted over the building blocks as just blobs of black and then added small stroke details for the lit windows and such

These backgrounds are traditionally made painting with poster colors where the artists will roughly lay out the color scheme over a sketch with very watered down color, then apply dryer and dryer color to bring in the details.
