Hi Jongmin, thanks! That's an interesting possibility. Based on the reported Polycomb abundance measurements, Polycomb protein occupancy seems likely too low to be able to block loop extrusion. It would be super interesting to look at E-P loops in cell lines +/- degrons to get more insights on this!

Genome structure mapping with high-resolution 3D genomics and deep learning
Gene expression is often regulated by distal enhancers through cell-type-specific 3D looping interactions, but comprehensive mapping of these interactions across cell types is experimentally intractab...
Excited to share my first preprint from the @andersshansen.bsky.social lab! It's got genomics, AI, and biology - all wrapped up in a great paper. Also very grateful to my amazing collaborator, Fan, for all his hard work. Please check it out and let me know what you think!

Q: How do we get ultra-high-res 3D genome maps?
A: New deep learning model, Cleopatra.
Cleo trains on Micro-C, fine-tunes on RCMC, and predicts genome-wide 3D maps at ultra-high resolution.

Super excited to share my first preprint from @andersshansen.bsky.social Lab! We used MINFLUX to track chromatin (H2B-Halo and Fbn2 locus) at an unprecedented 200β―ΞΌs, then combined it with SPT to span ΞΌs-minutes (H2B) or SPT & Super-Res Live-Cell Imaging (SRLCI) to span ΞΌs-hours (Fbn2)
