Extracellular Vesicles Define Discrete Nano‐Based Niches Within the Human Haematopoietic System Stem cell niches are complex multi-signalling networks comprised of molecular cues and physical interactions, orchestrated by niche-resident cells and the extracellular factors they produce. The bone....

Enriching for Extracellular Vesicles from Human Bone Extracellular vesicles (EVs) are nano-sized membrane-bound structures thought to be secreted by all cells and increasingly recognized as key mediators of intercellular communication. Established EV isolation protocols for bodily fluids-primarily focus on blood with limited insights into methods optimized for EVs from other hematopoietic regions. In this study, we present a novel protocol for the isolation and enrichment of EVs from human trabecular bone and bone marrow. This method employs a two-step purification strategy, combining iodixanol density cushion (IDC) ultracentrifugation with size exclusion chromatography (SEC), and enables EV recovery from fresh tissue hours after collection. Importantly, this approach facilitates the enrichment of bone-derived EVs without the need for enzymatic digestion or long-term culture, preserving native EV populations. This protocol offers a valuable tool for researchers investigating EVs derived from the diverse cellular constituents of the bone microenvironment. ### Competing Interest Statement The authors have declared no competing interest. I think people both within science and outside science don’t fully appreciate the miracle that were the Covid-19 vaccines. How much courage it took to get the sequence out, how much work had already gone into figuring out the best viral antigen and how best to present it. 1/

Anyone know of a site for posting excuses from students to re-do exams/assignments? Its exhausting being inundated with requests for extra marks to the detriment of other students. I won't even start with the 30% accommodations that already exist. I fear for the next generation workforce..

Blebbisomes are large, organelle-rich extracellular vesicles with cell-like properties - Nature Cell Biology Jeppesen, Sanchez et al. identify blebbisomes as large membrane-enclosed vesicles containing organelles and secreted by cells. Blebbisomes can both secrete and internalize exosomes and microvesicles.

Two big reveals in the world of #extracellularvesicles this week:

Post image “But there is another world that is also disturbing me,” Cage continued. “It’s happening right now around all of us: the new AI world. I am a big believer in not letting robots dream for us. Robots cannot reflect the human condition for us. That is a dead end if an actor lets one AI robot manipulate his or her performance even a little bit, an inch will eventually become a mile and all integrity, purity and truth of art will be replaced by financial interests only. We can’t let that happen.
