In 2026 if we need a good spectral library for a weird tough sample do we still run the HPH fractions with DDA? That was my default for compatibility with more tools, but I’d appreciate other opinions.

I'm continually surprised when I just plug a PTM into modern global DDA proteomics data search and - boom - there's a pile of peptides with that PTM that make sense. I worry students will miss out hand rocking 4kg of peptides with $30k in mABs in a walk in 4C for 20 hours to find 3 acetylations.

The TMT on a TimsTOF paper was rejected 6 times. Half of those were because “MS3 is necessary for good quan.” The other half was because the first author is a mediocre scientist and writer. He was worse then.

I handed in my keys to my labs to Johns Hopkins at 2pm and my email was GONE by 8am the next day. It was life altering but it reinforced that leaving was the right decision. Program officers love their emails bouncing. It makes a place seem kind, organized and professional.
