Differential pathological dynamics triggered by distinct Parkinson patientβderived Ξ±-synuclein extracts in nonhuman primates
Distinct Ξ±-synuclein assemblies trigger distinct neurotoxic cellular pathways.
Iβm very happy π₯³ to share our new collaborative work, out today in Science Advances! Many thanks to all the co-authors!

Maintenant : abrogation.

The ground truth for AD is a bit of a debate recently, to say the least! But I am a big fan of the intersection between a clinical diagnosis and a biological definition.

- we need to learn from system biology regarding the interpretation of gene hits. For too long we have relied on imprecise tools (gene ontology) that are often wrongly annotated and cannot sufficiently account for the redundancy of biological systems or the existence of feedback loops

Interesting read! Some thoughts: - a GWAS is only as good as the patient inclusion. There has been immense efforts for AD on that aspect, even doing small postmortem GWAS on path confirmed cases
- the absence of a coherent picture at the gene level might indicate multiple disease mechanisms

In vivo hyperphosphorylation of tau is associated with synaptic loss and behavioral abnormalities in the absence of tau seeds - Nature Neuroscience
Mice expressing humanized mutant tau exhibit synaptic loss and behavioral abnormalities in the absence of abnormal tau conformers, suggesting that hyperphosphorylated tau can be pathological, at least...
Paper alert from the Duff Lab π¨π¨ My amazing colleagues showed that, in novel MAPTKI models, hyperphosphorylation (in absence of aggregation or even seeding) is sufficient to drive synapse loss and cognitive deficits. Huge implications for preclinical research and the way to therapies. Have a read!!
