Congrats to first author Venda for putting it all together! This project was a great collaboration with the @titiasixma.bsky.social lab who helped us give structural meaning to our observations.
Kaima Tsukuda and @andrewblackford.bsky.social have a highly complementary study that indicates that mutations in DDIAS (and CIP2A) caused neurodevelopmental defects, pointing a role for this pathway in development. /fin

Why is an ssDNA-binding protein involved in the CIP2A pathway? Our data suggests it may be to prevent ssDNA from accumulating in mitosis.
We propose that mitotic ssDNA is a liability for the integrity of chromosomes during cell division, and that a function of CIP2A-TOPBP1-DDIAS is suppress it.
We were puzzled by the fact that DDIAS has an OB-fold domain which often acts as an ssDNA-binding domain.
We find that DDIAS binds to ssDNA. This activity is not involved in its recruitment to DNA damage sites, but it is necessary for its function in genome maintenance.
Like CIP2A, DDIAS accumulates at DNA lesions specifically during mitosis, and it is recruited to the CIP2A-TOPBP1 complex via a phosho-dependent interaction with the TOPBP1 C-terminus.
DDIAS loss, like that of CIP2A, impairs the viability of BRCA1/2-deficient cells.
