Insights: Frosio and Ostuni @unisr.bsky.social discuss new study by Gorin et al. (rupress.org/jem/article/...) which reveals an unexpected mechanism by which IFN-γ sustains trained immune states through prolonged signaling driven by cytokine retention at the cell surface. rupress.org/jem/article/...

Translational research is digging for gold. Basic research is learning where and what to dig. The more funding is diverted to translation, the more we deplete the research ecosystem, driving the supplies of "gold" - immediately translatable findings and innovations - to unsustainably low levels.

IL-1β+ macrophages and the control of pathogenic inflammation in cancer While highlighting the complexity and heterogeneity of tumor immune microenvironments, the application of single-cell analyses in human cancers has identified recurrent subsets of tumor-associated macrophages (TAMs). Among these, interleukin (IL)-1β+ TAMs – cells with high levels of expression of inflammatory response and tissue repair genes, but with limited capacity to stimulate cytotoxic immunity – are emerging as key drivers of pathogenic inflammation in cancer. In this review we discuss recent literature defining the phenotypical, molecular, and functional properties of IL-1β+ TAMs, as well as their temporal dynamics and spatial organization. Elucidating the biology of these cells across tumor initiation, progression, metastasis, and therapy could inform the design and interpretation of clinical trials targeting IL-1β and/or other inflammatory factors in cancer immunotherapy.

Happy to share our reflections @cp-trendsimmuno.bsky.social on our favourite cells: IL-1β+ macrophages 🔥🔥🔥
Molecular, spatial, temporal, functional properties in cancer and beyond... it's all in there from the awesome Nicoletta Caronni, Federica La Terza and @frosio.bsky.social

Last week in Vancouver was wild—my first time overseas didn’t disappoint ❄️. Got chills sharing our work on IFN regulation and seeing it finally go international 🦠. So thankful to the organizers and to who is making the dream possible @ostunilab.bsky.social @keystonesymposia.bsky.social
