3/ We compared the damage profile with mutational hotspots and heteroplasmic positions and found that 2,781 positions had not been previously reported in any of those profiles.

Distribution of the 2933 hotspots identified in the present study. These hotspots exhibit a percentage exceeding 23.8% and collectively represent the top 25% of the positions with the highest values. The non-coding region is highlighted in red, while an orange line denotes the origin of replication. Region of the mitogenome with < 30 reads covering them in at least 75% (321 samples) of those analysed are shaded in blue to indicate their absence in our profile.
2/ Using 427 non-UDG-treated ancient samples from NGS data, we created a comprehensive molecular damage profile for ancient mtDNA, identifying 2,933 damage hotspots across the human mitogenome (out of 11,735 evaluated positions).
