Splicing of a core clock gene regulates seasonal adaptations by a winter gating mechanism Seasons bring changes to the environment. Many organisms adjust their physiology and behavior in response to seasonal changes in order to survive. Although the molecular mechanisms mediating the integ...

Thrilled to share our new work! In this study, we have expanded on the role of the circadian clock in integrating seasonal environmental cues through splicing.
Thanks to @joanna-chiu.bsky.social for the continuous mentorship and to all co-authors!

4. O-GlcNAcylation also interacts with phosphorylation. (They both modify serine, threonine and tyrosine.)

3. Rhythmic O-GlcNAcylation directly modulates proteins involved in gene expression.

2. This rhythm is responsive to changes in mealtime. (So glad that TMT proteomics worked out for this PTM!)

1. Nuclear protein O-GlcNAcylation shows daily rhythms in mouse liver.

These led us to explore whether proteome-wide O-GlcNAcylation mediates the effect of mealtime in rhythmic liver gene expression. We found:

We started out asking how mealtime regulates genome-wide rhythmic gene expression. We knew several nutrient-sensitive pathways are involved. We knew O-GlcNAcylation, a nutrient-sensitive protein modification, modulates CLOCK, BMAL1 and PER2.

O-GlcNAcylation of nuclear proteins in the mouse liver exhibit daily oscillations that are influenced by meal timing The liver transcriptome is highly sensitive to metabolic cues. By characterizing nuclear protein O-GlcNAcylation in the mouse liver over the course of the day and in response to altered meal timing, t...

Our paper is published in PLOS Biology! journals.plos.org/plosbiology/.... What a long and rewarding journey with @joanna-chiu.bsky.social and many who are not on Bluesky.
