The interest here is exploratory where the subunits in complex are uncharacterized..

Peptide:TMT ratio and pH are major factors influencing labeling efficiency.. Ensure both variables are controlled.. Could inject a labeled sample before pooling to quantity labeling efficiency.. Use TMT as a dynamic mod for this.. ideally up to 98% labeling on K and ~95% on N-term is anticipated..

I am a fan of TMT. There is no reason to feel intimidated. It will be alright. Follow the protocol. Design the experiment carefully and follow the steps diligently. Ensure normalisations and labeling efficiencies are checked and controlled. The TMT quantitative data is awesome. All the very best.

Guess at which point I realized that the column heater is not connected.. I am very glad that this is the only boss-back-on-bench catastrophe so far.. The muscle memory from years of proteomics works still.. Thankfully, nothing got blown up.. yet.. 😂😂

Changed the 50um lines to 20um.. With this setup the delay is not larger than what generally is for a setup where the column oven is in use (like when uPAC columns are being in use).. Frankly, the plan is to set it up as a single neo instead of a duo for now.. Yet to test this setup though..
