This study presents a novel biomimetic nanoparticle platformβ€”Caco-2 cell membrane-coated, manganese-doped ZnO nanorodsβ€”for targeted microRNA (miR-200c-3p) delivery to intestinal epithelial cells. By combining magnetic responsiveness, homotypic targeting, and lipid membrane camouflage, the engineered nanoparticles achieve site-specific delivery, enhance intestinal permeability, and modulate tight junction integrity without inducing toxicity.

The work demonstrates, for the first time, that:

miRNA-loaded nanorods can be guided magnetically and penetrate the mucus barrier.
Homotypic targeting using epithelial cell membranes improves localization and uptake.
miR-200c delivery via this platform significantly alters occludin expression and gut permeability in vivo.
This approach opens new avenues for precision therapeutics in IBD and barrier dysfunction disorders, offering a biocompatible, traceable, and customizable delivery system for gut-targeted RNA therapies.
