#EU law requires #GMO labeling when levels exceed 0.9% at the ingredient level.

Our study shows that duplex #sequencing can reliably detect #NGT genome-edited DNA down to 0.1%, suggesting a potential alternative detection approach when #PCR assay design is not feasible. ๐Ÿงฌ๐Ÿ”Ž

By using duplex sequencing, we managed to call all expected mutations and filter out PCR artifacts and sequencing errors. Despite the excellent detection performance, challenges remain with respect to accurate quantification.

Yet, when PCR assays are unfeasible, sequencing can be a useful tool! 7/

Detection performance was very good: we reliably detected all expected mutations down to 0.1% (if enough DNA was used). โœ…

Other unexpected mutations were found, but none was called in all replicates, and in both strands of at least two distinct DNA molecules. We could filter them out! โœ… 5/

To test it, we mixed non-modified #tomato ๐Ÿ… DNA with edited DNA at decreasing levels (from 10% down to 0.1%), and we sequenced three replicates for each level.

Enter #duplex sequencing: a clever technique that tags #DNA molecules with barcodes before PCR. It tracks both strands of each molecule, so you can discard the mutations that were present in only one molecule or in only one strand.

It's not easy to design #PCR assays that reliably distinguish sequence differences of just one nucleotide.

On the other hand, #sequencing struggles to reliably call low-frequency mutations. At those levels, noise is high and technical artifacts are a real issue. 2/

I discovered that 54% of my followers prefer to write in English, and only 42% prefer Italian (on #Mastodon it's the opposite: Italian 65%, English 31%).

From now on I'll write here in #English ๐Ÿ‡ฌ๐Ÿ‡ง, and on Mastodon in Italian. The topics too will be different, this account will be more "scientific".
