We will use genetic and pharmacological methods to confirm the oncogenic effect of candidate cancer drivers, and test strategies to reverse tumour growth.

Our plan is to use these organoids to understand the (epi)genetic underpinnings of medulloblastoma, a malignant childhood hindbrain cancer believed to arise during prenatal cerebellum development. /4

Most biomedical research is done in mouse models 🐭 , which do not adequately capture unique aspects of human πŸ‘ΆπŸΎ brain development, disease, or response to treatment. Organoids provide a much-needed human laboratory model for this purpose.

These are the result of reprogramming human skin cells into stem cells and then into simple, 3D models of the human cerebellum. The image shows cross-sections of the organoids expressing signature proteins of cerebellar neurons. e.g., SOX2 for neural stem cells, KIRREL2 for Purkinje cells. /2

Take a look at our first batch of human cerebellar #organoids! 🧠 🧫 Aren't they beautiful.
Collab between our lab + Liliana Attisano at U Toronto. Nice work Maria Mangos and Nishka Kishore from Pai Lab! Thank you Alex Atamian and @giorgiaquadrato.bsky.social group for guidance. #brainorganoids

Finally activated CoPilot on my VScode this morning. It's eerie, almost like it's reading my mind and I'm auto-completing a LOT ... feels like my long term risk of RSI is going down already... Hmm, let's see where the catch is...

Classification of non-TCGA cancer samples to TCGA molecular subtypes using compact feature sets Ellrott etΒ al. provide a means to assign patient samples from clinical trials and other cancer genome studies to published TCGA molecular subtypes. Applying machine learning to data from five different molecular platforms for 8,791 TCGA tumor samples, they contribute a public resource of 737 top classifier models, which can form the foundation for clinical assay development.

Thrilled to share the results in Cancer Cell of years of hard work to develop classifiers for all major TCGA cancer subtypes, enabling assessment of differential drug response among cancer subtypes in clinical trials. www.cell.com/cancer-cell/...

Finally a chance to process some astro data! Here is my image of beautiful Polaris / North Star / Dhruva ΰ€§ΰ₯ΰ€°ΰ₯ΰ€΅, surrounded by stars and interstellar dust (i.e., matter that doesn't emit light). Taken in October from our RASC observatory in the Blue Mountains. πŸ‘‡πŸΎ Gorgeous luminance detail below...

Congratulations to Ian Cheong, who successfully defended his MSc. thesis this afternoon! πŸ₯³ @mbpatuoft.bsky.social Ian's thesis is titled, "Human and mouse unipolar brush cells and relevance to Group 4 medulloblastoma". Also our first graduate from the Pai lab, so double milestone for us!
