The virus is shuttled to the nucleus, where (5) transcription of early virus genes is initiated, followed by (6) viral protein synthesis and (7) viral replication. (8, 9) The switch from early to late viral protein synthesis triggers the (10) assembly of new virions and (11) virus release. During infection, HAdV hijacks and activates the host cell machinery, including the activation of the PI3K/Akt/mammalian target of rapamycin (mTOR) pathway. The mTOR signalling is activated through two viral proteins, E4 ORF1 and E4-ORF4, which activate mTOR signalling directly (via PI3K) or indirectly (via PP2A), thus promoting cell growth, cellular and viral protein synthesis and potentially suppressing cellular antiviral response. Itraconazole interferes at two different stages of HAdV infection. The early antiviral effect of itraconazole is mediated by cholesterol sequestration in endolysosomes that hinders virus uncoating and thereby promotes endolysosomal degradation of HAdV. Additionally, itraconazole inhibits the virus-promoted activation of mTOR through activation of AMPK. AMPK inhibits mTOR signalling, thus affecting viral protein synthesis and survival of HAdV-infected cells (induces autophagy of infected cells to clear virus infection). Ku-63794 interferes with mTOR activation by directly inhibiting mTOR and by inhibiting Akt, a protein upstream of mTOR. Akt inhibition promotes TSC1/2 activation, resulting in Rheb inhibition and inactivation of the mTOR pathway. Both mechanisms contribute to lower virus particle production and decreased infection rates.
