I am a vision scientist interested in how our retina circuits process color and motion information.

9/In the HFseg1 volume, we annotated all Müller cells (555 cells, 92.2% of non neuronal cells) and small population of glial cells, astrocytes cells (32 cells, 5.3% of non neuronal cells) present in the GCL.

8/Synaptic detection:Together with Dr. Tommy Macrina and the Zetta AI team(zetta.ai), we combined deep learning with ground truth manual labeling to successfully detect all synapses and their synaptic partner relationships, as well as to perform alignment and segmentation in the HFseg1 volume.

7/Ganglion cells: We annotated 599 ganglion cells. The midget (536), parasol (26) and small bistratified (12) cells account for 95.8% of the ganglion cell in HFseg1. The remaining 25 ganglion cells show relatively large dendritic fields, various morphological forms.

6/Amacrine cells:. We identified 293 amacrine cells, including 40 cells displaced to the ganglion cell layer. Most amacrine cells showed cell type-specific nuclear staining (from strongly heterochromatic to strongly euchromatic) and nuclear shape (from smooth and round to deeply infolded).

5/We identified 912 bipolar cells in he HFseg1 volume, including the major types recognized in non-human primates, though with some notable differences.

4/Horizontal cells:We annotated 265 horizontal cells in the HFseg1 volume, and found that almost all (255 cells) were H1 cells, with a very small population of H2 cells (10). This significantly higher density of H1 cells relative to H2 cells altered their synaptic connectivity with S and LM cones.

3/Photoreceptors: Our HFseg1 volume contained 313 cone (pedicles) and 24 rod (spherules) synaptic terminals. Fifteen cone pedicles were identified as short wavelength sensitive (S) cones based on distinctive connectivity.

2/The fovea is a small, specialized region of the retina serving high acuity and color vision in humans and non-human primates. Foveal retinal cell transcriptomes are now available. But, how the transcriptome governs neural cell types and neural wiring diagrams remains poorly understood.

1/ We are excited to share our new manuscript. Here, we provide a nanoscale connectome of the human foveal retina. Our dataset represents the first connectome of any complete neural structure in the human nervous system.
