It evolved early and transformed the biosphere. But it evolved 2x. Why not just once, why not more? Our work suggests the answer is priority effects.

Trom top left to bottom right: SPY505-DNA (green); SPY555-DNA (magenta); SPY595-DNA (yellow); SPY620-DNA (orange); SPY650-DNA (red); SPY700-DNA (darker red). a white scale bar of 50 micrometers is given for each image to get an impression of the scale of each nuclei.

We have awesome news for you! All our 6 SPY-DNA probes work beautifully well with the U-ExM protocol.
You can now tune the color of the DNA channel to your needs, according to the fluorophores on your preferred antibodies.

Whatever leftover gels (those you don't stain), can be kept at 4Β°C in PBS + Azide 0.01% + BSA 2% for months.
You could even put them in multiple 50% glycerol washed and leave them at -20Β°C.
Even stained gels, if left in PBS+Azide could keep their signal for weeks.
One of the big pluses of ExM
wow - if you have enough cells on the gels, and wanna send us a spare gel piece, we can defnietly try some more antibodiess !
