We had already tried some Chem fixed samples with Kenneth, a couple years back without much success. I think going cryo would be needed here ! And yes, happy to try again

Priority effects inhibit the repeated evolution of phototrophy - npj Complexity npj Complexity - Priority effects inhibit the repeated evolution of phototrophy

New paper out! Here's a puzzle: phototrophy, the ability to use light for energy, is one of life's great innovations. It evolved early and transformed the biosphere. But it evolved 2x. Why not just once, why not more? Our work suggests the answer is priority effects.

I also very much enjoyed being in full handle of the #Zeiss 3Dsim scope provided generously for us to explore the various samples.

This one was quite the journey! The paper describing the #ChlamyDataset is finally out and on the cover of Mol Cell!

This beautiful rendering made by co-author @jessheebner.bsky.social and Holly Peterson shows an instance of mitochondrial fission found in the dataset 😍

[Maybe long thread ahead]

This is the cover proposal I made for our recent paper!

It’s Pyramimonas diskoicola β€” a very beautiful green flagellate and one of those cells that makes you stop and stare for a while.

I tried to capture its shape, symmetry, and general vibe :)

The images shows an array of 6 Expansion microscopy images of cell nuclei (three columns and 2 rows). each image was taken with a different SPY-DNA probe to stain the nuclear DNA and is pseudocolored to the fluorescence wavelength of the SPY-DNA probe. Trom top left to bottom right: SPY505-DNA (green); SPY555-DNA (magenta); SPY595-DNA (yellow); SPY620-DNA (orange); SPY650-DNA (red); SPY700-DNA (darker red). a white scale bar of 50 micrometers is given for each image to get an impression of the scale of each nuclei.

Are you staining DNA in your ExM samples?

We have awesome news for you! All our 6 SPY-DNA probes work beautifully well with the U-ExM protocol.

You can now tune the color of the DNA channel to your needs, according to the fluorophores on your preferred antibodies.

Whatever leftover gels (those you don't stain), can be kept at 4Β°C in PBS + Azide 0.01% + BSA 2% for months.
You could even put them in multiple 50% glycerol washed and leave them at -20Β°C.
Even stained gels, if left in PBS+Azide could keep their signal for weeks.
One of the big pluses of ExM
