Re-adenylation by TENT5A enhances efficacy of SARS-CoV-2 mRNA vaccines | Nature Despite the widespread use of mRNA vaccines against COVID-19, little is known about the metabolism of therapeutic RNAs. Here we use nanopore sequencing1–3 to analyse individual therapeutic mRNA molecules, focusing on their poly(A) tails. We show that the Moderna mRNA-1273 vaccine4 has a poly(A) tail of around 100 nucleotides, followed by an mΨCmΨAG sequence. In cell lines, mRNA-1273 undergoes rapid degradation initiated by mΨCmΨAG removal, followed by CCR4–NOT-mediated deadenylation. However, in medically relevant preclinical models, particularly in macrophages, mRNA-1273 poly(A) tails are extended to up to 200 nucleotides by the TENT5A poly(A) polymerase5–7, which is induced by the vaccine. Re-adenylation, which stabilizes target mRNAs, is consistently observed in synthetic mRNAs that encode proteins targeted to the endoplasmic reticulum, such as ovalbumin or antigens from Zika virus8 or the malaria parasite9. Welcome folks!!! This group of volunteers are amazing. I might be biased.

"My first HG pregnancy was horrific. I couldn't access Cariban at first, and then I couldn't afford it. My daughter stopped growing and was born tiny, and I was left with several health problems afterwards. It took years for me to feel like I was ready to get pregnant again.

This time I was given medication by my GP to take preemptively and then Cariban at 6 weeks. Even at its worst, it has been easier than the first time. And this baby is measuring right where she should be. My second pregnancy (although still very difficult) has been a night and day difference to my first, and has been very healing for me.

This is why we exist. This is the difference that early and proactive treatment can make to HG patients and their babies. Thanks to Trish in our support group for sharing with us.

How our democracy has been hijacked by corporations and wealthy elites The only effective remedy is to ban all private donations, or bribes, to political parties.

Political donations are institutionalized bribery.

Donors get access to policymakers, shape laws, tax privileges, lax regulations, govt contracts/subsidies, honours, power.

Banning political donations is a necessary step for free and fair elections and democracy
