Fun little tidbit from the paper: In hydroamination of substrate 2p to give 4p, the equilibrium constant between N-oxide and N-OH is only 0.056; but the addition of Os catalyst to selectively reduce the N-oxide shifts the equilibrium towards N-oxide, and translates into 83% isolated yield!

Enabling Unfavorable Hydroamination Reactions Using a Chemoselective N–O Bond Reduction Despite major advances, intramolecular alkene hydroamination reactions often face limitations. Herein, a redox-enabled process featuring oxidation of an amine to a hydroxylamine, a concerted hydroamination step, followed by catalytic reduction of N-oxide is shown to be broadly applicable. Catalyst screening and optimization showed that a K2OsO2(OH)4–pinacol complex rapidly and chemoselectively reduces the N-oxide cycloadduct in the presence of hydroxylamine and dimethyl sulfoxide. This selectivity was exploited to drive the equilibria toward complex products.

The Beauchemin Group was delighted to publish this work in late 2024. PhD student Huy Ly, with undergrad Hala Almeneim, did incredible work! Thanks to Jeff Keillor for support in kinetic analysis.
