The beautiful thing is, we provided genetic diagnosis using EXISTING sequence data - meaning patients with NCF1-CGD lacking a genetic diagnosis, and their at-risk relatives can now know their mutation and risk status.

We masked the pseudogenes and aligned all reads to NCF1, allowing variant calling. (Fig 2A). Using 1000G as a reference, we show that most NCF1-CGD patients carry 6 copies of the 2-base deletion (Fig 2B) and the variant frequency (AltAB) of that is in concordance with ddPCR data (fig 2C)

Excited to share our new paper identifying NCF1 mutations in chronic granulomatous disease (CGD) in 47/48 pateints. NCF1 diagnosis is impacted by gene duplication resulting in 2 pseudogenes each of which delete 2 bases causing frameshift.
