That is a lot of cash burned! Looks like the only thing they are moving forward with is a molecule that makes beta-catenin form condensates. This seems completely opposite to the vision statement of the company

Polo kinase inhibits protein phosphatase 1 to promote the spindle assembly checkpoint and prevent aneuploidy Protein phosphatase 1 (PP1) is essential for spindle assembly checkpoint (SAC) silencing and mitotic exit, but its regulation during mitosis remains iโ€ฆ

Our latest research in collaboration with @nilssonlab.bsky.social, @andrea-musacchio.bsky.social and Mathieu Bollen reveals a new regulatory strategy employed by cells to control PP1 in mitosis. Check it out in @currentbiology.bsky.social

Nice. Intriguing with UBE2C - would think the protein is unstable so you would slow down APC/C and prolong arrest.

SLiM team resources @ ICR

In addition to uncovering CYB5R4 our functional map hints at many additional regulatory pathways and mechanisms that can now be explored by the community. Thanks to Norman Davey this will be facilitated through a website allowing exploration of the base editor screens slim.icr.ac.uk/base_editing...

16.02.2025 13:45 โ€” ๐Ÿ‘ 0    ๐Ÿ” 0    ๐Ÿ’ฌ 1    ๐Ÿ“Œ 0

We find that CYB5R4 mediated activation of PP4 is critical for controlling the response to the DNA damaging agents cisplatin and Illudin S that cause elevated levels of ROS. This extends earlier important observations from the @durocher1.bsky.social lab on CYB5R4 (Olivieri et al 2020).

16.02.2025 13:45 โ€” ๐Ÿ‘ 1    ๐Ÿ” 0    ๐Ÿ’ฌ 1    ๐Ÿ“Œ 0

We uncover CYB5R4 as an activator of PP4 and PP6 but not the closely related PP2A. CYB5R4 is a dedicated reductase for the catalytic metal ions and we show that this pathway is conserved from yeast to man. Thus PP4 and PP6 activity is regulated by oxidation and reduction of the metal ions.

Happy to share our latest work on phosphoprotein phosphatase (PPP) regulation. We integrate CRISPR knockout and focused base editor screens to uncover the regulatory landscape of PPPs (more below). Great collaboration with the Zhu, Davey, Kettenbach and Ogris labs.
