SDH inhibition elevates succinate, inducing SHMT2 succinylation that suppresses mitochondrial one-carbon flow and formate supply, thereby constraining purine assembly. Cancer cells compensate via purine salvage, and dual inhibition of SDH and salvage yields significant antiproliferative effects and suppresses tumor growth.
