This work defines new rules of miRNA-mRNA engagement, and demonstrates the value of fine-scale molecular engineering in studying gene regulation.

🔬 Key findings:

1. Pairing beyond the seed significantly enhances miRNA repression for select targets, which we define.

2. Structural features like GU wobble positions and bulge orientation affect binding.

3. Abasic modifications offer a precise new tool for dissecting miRNA-mRNA interactions.

MicroRNAs target genes through their “seed” region — but the contribution of other regions has been less clear. We used a novel molecular approach: abasic nucleotide modifications to disrupt pairing with surgical precision, and assessed gene regulatory impact at RNA and protein levels in tandem.
