Yeah I spent weeks just like gazing at gene of interest sequences trying to manifest the best hypothetical intron/exon boundaries and like there has to be a better way

Also a lot of fun describing “weird shit upstream/downstream of highly expressed sequences with no real discernible strong promoter characteristics” for possible IP reasons

Kinda maddening because all experiments were basically built on this assumption that we’d just be able to use everyone’s favorite promoter and get great activity and that was just never the case

Back when did promoter screening we found little correlation with reported promoter activity but if you were willing to screen like a dozen candidate promoters, 3’ UTRs, and first exons we always found something that worked great but again basically no predictability
