Other complex III inhibitors are being developed as drug leads for apicomplexan diseases.
Together with Stone Doggett and colleagues at @OHSUNewswe we showed how the drug-lead ELQ-300 binds Toxoplasma complex III and designed a more potent version.
Atovaquone is a clinically used drug for apicomplexan diseases such as #malaria and #toxoplasmosis, but its precise mechanism of inhibition was unknown.
We showed how atovaquone interacts with Toxoplasma complex III and why it is selective to apicomplexan compared to human.
Respiratory complexes form βsuper-complexesβ whose physiological function is debated, and assembly only studied in a few organisms.
We showed that apicomplexan conserved proteins mediate III2-IV supercomplex formation, and that this provides competitive growth advantage.
Parasites need mitochondrial respiration to infect and spread. Effective anti-malaria and ani-toxoplasmosis drugs target respiratory complexes (e.g. atovaquone)!
But how do the parasite complexes work and how do drugs inhibit them?
