Sex isn’t a nuisance variable.
It’s not noise.
It’s biology.
Excluding it doesn’t simplify the story, it hides it.

Including both sexes didn’t complicate our design.
With a factorial structure, power is preserved, even with fewer mice per group.
It’s efficient, rigorous, and it’s how we found the signal.

FGF21 was known to rise with fasting or amino acid imbalance.
But a sex- and context-specific developmental role under physiological protein restriction?
That’s novel.

Using liver-specific knockouts, we tested causality:
❌ Deleting hepatic Fgf21 led to partial rescue of growth and earlier puberty in females.
✅ In males, no effect.
FGF21 acts only in females, and only when protein is limited.

We then identified a key mediator: FGF21, a liver-derived hormone.
Under protein restriction, it rises in both sexes, but only delays growth and puberty in females.

Males entered puberty on time, despite stunting.
Females, in contrast, showed delayed vaginal opening and estrus onset.
Same stress, different developmental trajectories. The timing of puberty is more sensitive in females.

Males: severe and persistent stunting.
Females: transient early impairment, but mostly recovered by adolescence.
Same diet, different outcomes. The biological response to protein restriction is profoundly sex-specific.

We assumed sex wouldn’t matter. We were wrong.
By including both males and females, Amélie uncovered hidden sex-specific strategies in response to early-life protein restriction.
