The Bill & Melinda Gates Foundation funding for this hugely collaborative project brought together polymer chemistry and microfabrication expertise in the Stevens group with the immunology insight of the @robinshattock.bsky.social group, most of whom are not on Bluesky (yet)!

We found that SCRIBE-mediated sustained OVA release profiles correlated with higher antibody titers and subtype class switching. By delivering model vaccine boosters with delayed first order kinetics, we induced equivalent titers to those of a double injection formulation.

Inspired by the osmotic pump, the porthole in our hollow microparticle allowed for a precise investigation of how resin thickness and chemistry control protein release kinetics. We were also able to maximize injectability with spherical designs that are inaccessible to other microfab strategies.

To the best of our knowledge, we achieved the highest resolution printing with PLGA yet (500 nm XY, 1 ΞΌm Z) with printing error < 150 nm. The key breakthrough for me was realizing that we could blend various comonomers with the PLGA-TA to tune the print properties without compromising resolution.

We introduce a new family of PLGA-based resins for two-photon polymerization (2PP) 3D printing, Spatiotemporal Controlled Release Inks of Biocompatible polyEsters (SCRIBE), and show how top-down microparticle design can tune immune responses to model subunit vaccines.
