How do SMC complexes load onto DNA to get ready for loop extrusion?
@roisnehamelinf.bsky.social & co discovered that Wadjet, an SMC complex involved in bacterial DNA immunity, performs some impressive molecular gymnastics π€ΈββοΈπ€ΈββοΈπ€ΈββοΈ.

I'm very pleased to share this work, which discovers a new material property of DNAβthe bending frictionβ in essence how "sticky or viscous" DNA is as it bends & loops due to all the complex molecular interactions within itself & water

Very pleased to share our new story on SMC complexes, a great team effort from @roisnehamelinf.bsky.social, @taschner.bsky.social in the GruberLab and James of @dnatopologylab.bsky.social We find that SMC hinge domains are ssDNA gates to facilitate obstacle bypass during DNA loop extrusion!

Graphical abstract showing the loop extrusion mechanism of the Wadjet complex, which selectively cleaves circular DNA while leaving linear DNA unaffected.
Happy to see our research on SMC Wadjet published in Molecular Cell. We showed DNA loop extrusion (LE) by a bacterial SMC, confirming LE is conserved across all life forms. Wadjet's LE activity is crucial for recognizing and restricting foreign plasmids, serving as a defense mechanism. π§¬π¬π§ͺ
