Yeah I end up just dipping individual bud clusters into a 2mL round bottom tube with 1mL of agro in that exact buffer. I felt the whole plant dipping to be not only wasteful but likely inducing some immune or osmotic response. silwet is very strong at breaking surface tension...and so much mess! ty!

At this point im just collecting UV dosimeter cards. This one is both short and long wave UV with scale of intensity as saturation of fluorescence. I like the generous UVC indicator target on this new one. Fairly cheap and infinitely reusable. Works for UVA/B too for sunlight.

Variations in wall thickness and warping due to autoclaving makes repositioning the tube exactly as you did during blanking tricky. Easier to just shoot through the length of the clear tube and adjust sensor value to liquid volume. Its all relative readings anyway.

Progress on a simple 2mL round bottom tube spectrometer for direct OD600 readings. Sides of tube are often graduated causing weird variations and will alter signal if not perfectly aligned to blank position. Reading through top, though longer light path, is more uniform. Testing.
