Do other PARPs label substrates for degradation as well? In which conditions is this most relevant? How do ADP-ribose and ubiquitin connect to regulate degradation?

Q for the hive-mind ๐ง : What transfection reagents do people like for big constructs these days? (I am thinking #HSV encoding BACs, ~170 kb). We've recently had poor performance with Trans-IT LT1 and Lipofectamine 2000, our two go-tos. Target cells are U2OS. Suggestions or RTs greatly appreciated ๐

Reminder: Nobel-prize winning PCR (1983), used in basically all genetic tech today, was only possible because of extremophile bacterium discovered in 1964 in Yellowstone funded by a small ~$80k NSF grant with no obvious application at the time. #science ๐งช
