I'm grateful for Nonso's hard work on this project, Jรถrg's assistance with autophagy, Bernhards' hosting of our little group and the excellent teamwork with Roko.

The reviewers and editor from the Journal have also been great to shape the revisions and make the paper better.

Thank you all ๐Ÿ™

Do other PARPs label substrates for degradation as well? In which conditions is this most relevant? How do ADP-ribose and ubiquitin connect to regulate degradation?

Redirecting

This is a very exciting finding, as it connect well with several recent publications that indicate that proteins modified by PARP7 are degraded. For example: doi.org/10.1016/j.ce...
doi.org/0.1038/s4431...
doi.org/10.1101/2025...

It also leaves many questions to be answered in future:

PARP7 is a proteotoxic stress sensor that labels proteins for degradation | The EMBO Journal imageimageADP-ribosylation is thought to occur on specific sites to affect specific characteristics of target proteins. Here, it is found to play a more promiscuous role, with PARP7 labelling proteins for efficient degradation during proteotoxic ...

I'm very excited to be able to share the latest work from our group with you:

doi.org/10.1038/s443...

We find that PARP7 is stabilised following proteotoxic stress and ADP-ribosylates proteins to enable their degradation. This occurs in a ubiquitin-dependent manner.

Q for the hive-mind ๐Ÿง : What transfection reagents do people like for big constructs these days? (I am thinking #HSV encoding BACs, ~170 kb). How a discovery in Yellowstone National Park led to the development of PCR - Richmond Scientific A discovery in Yellowstone National Park led to the development of PCR, the gold-standard COVID-19 tests used to fight the global pandemic.

Reminder: Nobel-prize winning PCR (1983), used in basically all genetic tech today, was only possible because of extremophile bacterium discovered in 1964 in Yellowstone funded by a small ~$80k NSF grant with no obvious application at the time. #science ๐Ÿงช
