...sorry i did not read the original message carefully, but according to my highly scientific analysis if gfp molecules truly didn't bind to actin (and if stress fiber was creating a bump) then there should be less cytoplasmic gfp where F-actin is...not more (yes, the "F" is used to annoy you).

But why is actin the only thing that's bumpy? There are probably a hundred other things in the ROI that exist but are not stained (like MTs)...first things to do is repeat with nothing in red channel and sequence the "GFP". Also...we need greyscale images.
